Product Specific Information

The M5/114.15.2 monoclonal antibody reacts with mouse MHC class II (I-A and I-E) glycoproteins, detecting polymorphic determinants on B cells, monocytes, macrophages, dendritic cells, and activated T lymphocytes from specific H-2 haplotypes (b, d, q, p, r, u).

This antibody inhibits I-A-restricted T cell responses in certain haplotypes and is supplied with:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Mouse splenocytes (≤ 0.4 µg/test; 100 µL final volume; 10⁵–10⁸ cells/test)

Note:

• NovaFluor dyes are incompatible with DNA intercalating viability dyes (propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for compatibility.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
• Add 5 µL per sample to antibody cocktails before combining with cells.

Target Information

MHC Class II molecules are transmembrane glycoproteins expressed on professional antigen-presenting cells (macrophages, dendritic cells, B cells). They bind exogenous antigens for presentation to T cells. The antigen-binding groove between alpha and beta chains is open at both ends and accommodates peptides 15–24 amino acids long.

HLA and MHC antibodies are crucial in Immunopeptidomics for identifying and characterizing neoantigens via LC-MS/MS analysis.
