Product Specific Information

The MEL-14 monoclonal antibody reacts with mouse CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells. It binds glycosylated, fucosylated, and sialylated glycoproteins such as CD34, glycam-1, and MAdCam-1, mediating lymphocyte rolling and homing to high endothelial venules (HEV) of peripheral lymphoid tissues.

Applications Reported: Flow cytometric analysis
Applications Tested: Tested by flow cytometric analysis of mouse splenocytes. Recommended use ≤0.125 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best performance.

Fluorochrome Information:
Brilliant Violet™ 480 (BV480) emits at 479 nm and is excited by a 405 nm violet laser. Ensure instrument compatibility.

Staining Recommendations:
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize non-specific interactions.

Fixation Compatibility:
Brilliant Violet™ 480 (BV480) is not compatible with methanol-based fixation.

Target Information

Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein adhesion receptor composed of:

  • C-type lectin binding domain
  • EGF-like domain
  • Two short consensus repeat (SCR) sequences
  • Spacer region, transmembrane region, and cytoplasmic tail

It is constitutively expressed on most leukocytes, including lymphocytes (excluding many memory T cells), monocytes, and polymorphonuclear cells.
