Target Information

Aggrecan, also known as cartilage-specific proteoglycan core protein (CSPCP) or chondroitin sulfate proteoglycan 1, is encoded by the ACAN gene.
It is a key component of the extracellular matrix in cartilaginous tissue and provides resistance to compression in cartilage.
The human protein is approximately 2316 amino acids (~250 kDa) and exists in multiple isoforms due to alternative splicing.

Aggrecan has a bottlebrush-like structure with chondroitin sulfate and keratan sulfate chains attached to a protein core.
Its synthesis and degradation are closely studied in relation to cartilage deterioration during joint injury, disease, and aging.
It contains three interglobular domains (G1, G2, G3) involved in aggregation, hyaluronan binding, cell adhesion, and chondrocyte apoptosis.

Aggrecan fragments are released into synovial fluid during all stages of human osteoarthritis due to cleavage by aggrecanase and MMP activity.
Antibodies BC-3 and BC-4 can detect neo-epitopes of Aggrecanase (ARGSVXXX) and MMP (XXXVDIPEN), respectively.
