This antibody is predicted to react with mouse and rat based on sequence homology.
Heat-mediated antigen retrieval is recommended prior to staining, using a 10 mM citrate buffer (pH 6.0) for 10 minutes, followed by cooling at room temperature for 20 minutes.
After antigen retrieval, incubate samples with primary antibody for 30 minutes at room temperature.
A suggested positive control is breast carcinoma or mantle cell lymphoma.

Target Information

Cyclin D1 (PRAD-1, bcl-1) is a key cell cycle regulator that functions with CDK4 and/or CDK6 to phosphorylate the Rb protein. It is a proto-oncogene overexpressed in various human neoplasms, including mantle cell lymphoma (MCL). Cyclin D1 regulates protein phosphorylation, epithelial cell proliferation, and adipocyte differentiation.

The CCND1 gene encoding Cyclin D1 is located on chromosome 11. Cyclin D1 is synthesized during G1 phase and forms complexes with CDK4 or CDK6 in response to growth factor stimulation. These complexes inactivate the Rb protein and regulate CDK inhibitors such as p21Cip1 and p27Kip1. Without growth factor stimulation, Cyclin D1 is unstable and undergoes ubiquitin-mediated degradation, leading to G1/S phase arrest.

Cyclin D1 belongs to the highly conserved cyclin family, which regulates CDK kinases and coordinates mitotic events. Overexpression or mutation of Cyclin D1 contributes to tumorigenesis and makes it a valuable target for studies in cell cycle biology and cancer research.
