Product Specific Information

The monoclonal antibody 42D1 recognizes the CD85d (ILT4, LIR-2) protein, a member of the ILT/LIR/MIR receptor family.
CD85d is a 110 kDa single transmembrane receptor with three Ig-like domains and a cytoplasmic domain containing ITIMs (immunoreceptor tyrosine-based inhibitory motifs). Expression is limited to monocytes, macrophages, and dendritic cells. Its ligand includes non-classical class I HLA-G and -B molecules. Binding triggers interaction with SHP-1 phosphatase, contributing to immune tolerance during fetal implantation and possibly influencing organ allograft rejection.

Crosslinking of CD85d with 42D1 antibody inhibits calcium flux and enhances HLA-G tetramer staining, restoring the stimulatory activity of tolerogenic dendritic cells.

Each product contains:

  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported

This antibody is reported for use in flow cytometric analysis.

Applications Tested

Pre-titrated and validated by flow cytometry using normal human peripheral blood cells.
Recommended use: 5 µL (1 µg) per test (100 µL final volume).
Cell density: 10⁵–10⁸ cells/test.

Important Notes

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
  • Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
    • Add 5 µL per sample to antibody cocktails before combining with cells.
    • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on nucleic acid dye structures engineered for optimized fluorescent signatures with minimal spillover and spread.

Multiple transcript variants encoding distinct isoforms are known.
