  • Cellular Localization: Nucleus.
  • Specificity Comments:
    Recognizes a serine/threonine protein kinase required for checkpoint control mediating cell cycle arrest, activation of DNA repair, and apoptosis.
    In response to DNA damage and replication blocks, cell cycle progression halts through regulation of key cell cycle proteins.
    The encoded protein acts as a checkpoint regulator and putative tumor suppressor, containing a forkhead-associated domain essential for activation after DNA damage.
    When activated, it inhibits CDC25C phosphatase, preventing mitotic entry, and stabilizes p53, leading to G1 arrest.
    Also interacts with and phosphorylates BRCA1, aiding survival after DNA damage.
    Mutations are linked to Li-Fraumeni syndrome and may predispose to sarcomas, breast cancer, and brain tumors.

Target Information

CHEK2 (CHK2) is a serine/threonine kinase involved in the DNA damage checkpoint pathway.
Mutations in CHK2 are associated with cancer susceptibility.
CHK2 phosphorylates several modulators of cell cycle control, including tumor suppressor proteins such as p53 and BRCA1, contributing to DNA repair and cell survival.
It functions as a putative tumor suppressor, with mutations linked to Li-Fraumeni syndrome and predisposition to various cancers including sarcoma, breast, and brain tumors.
