Target Information
Anaplastic Lymphoma Receptor Tyrosine Kinase (ALK) belongs to the insulin receptor superfamily and is vital for brain development. Mutations, rearrangements, and amplifications in the ALK gene are associated with tumors such as anaplastic large cell lymphoma, neuroblastoma, and non-small cell lung cancer. The most common alteration is chromosomal rearrangement, creating a fusion gene between ALK (chromosome 2) and NPM (chromosome 5). The NPM-ALK fusion protein is oncogenic. ALK encodes a receptor protein-tyrosine kinase with a transmembrane and extracellular domain, absent in the transforming NPM-ALK product. ALK shows strong sequence similarity to LTK and plays a crucial role in neuronal development.
