Target Information
Amyloid beta peptide (Abeta/Beta-amyloid)는 알츠하이머병 환자 뇌의 아밀로이드 플라크 주요 구성 성분입니다. Abeta는 40–43개의 아미노산으로 이루어져 있으며, beta-amyloid precursor protein (beta APP)에서 두 단계의 절단 과정을 통해 생성됩니다.
첫 번째 절단은 beta-secretase (BACE)에 의해 이루어지며, 이는 prohormone convertase인 furin에 의해 활성화됩니다. 이후 gamma-secretase 복합체가 beta APP의 막내 절단을 수행하여 amyloid beta peptide를 생성합니다.
Gamma-secretase는 presenilin-1, presenilin-2, nicastrin 등을 포함하는 다서브유닛 복합체로 알려져 있습니다. Abeta는 신경섬유 엉킴(neurofibrillary tangles)과 아밀로이드 코어의 주요 구성 요소이며, Alzheimer’s disease, Lewy body dementia, Down’s syndrome, amyloidosis (Dutch type), cerebroarterial amyloidosis, Guam Parkinson-Dementia complex 등과 관련되어 있습니다.
