Target Information
Amyloid beta peptide (Abeta/Beta-amyloid)은 알츠하이머병 환자의 뇌에서 발견되는 아밀로이드 플라크의 주요 구성 성분입니다. Abeta는 40–43개의 아미노산으로 구성되며, 베타-아밀로이드 전구체 단백질(beta APP)로부터 두 단계의 절단 과정에서 생성됩니다. 첫 번째 절단은 베타-secretase(BACE)에 의해 수행되며, 두 번째 절단은 감마-secretase 복합체에 의해 이루어집니다. 감마-secretase는 presenilin-1 및 -2를 포함한 다중 서브유닛 복합체로 구성되며, nicastrin 단백질이 beta APP의 카복실말단에 결합하여 Abeta 생성 조절에 관여합니다.
Abeta는 신경섬유 엉킴 및 아밀로이드 코어의 구성 요소로서 알츠하이머병, 루이소체 치매, 다운증후군, 아밀로이드증(Dutch type), 뇌혈관 아밀로이드증(cerebral amyloid angiopathy), Guam Parkinson-Dementia complex 등 다양한 신경퇴행성 질환과 관련이 있습니다.
