Product Specific Information

eBioTY/11.8 recognizes CD73, a 69-kDa GPI-anchored cell-surface protein with ecto-5'-nucleotidase activity.
In mice, CD73 expression on myeloid cells (CD11b) is restricted to bone marrow. In humans, CD73 can be induced to secrete a soluble form with IL-2, suggesting a role in activation signaling.

CD73 is expressed on subsets of lymphocytes and increases during lymphocyte maturation. It is also found on memory CD4 T cells and regulatory T cells, suggesting roles in immune regulation.

Applications Reported:
This antibody is suitable for flow cytometric analysis.
Applications Tested:
Validated in flow cytometric analysis of mouse splenocytes; ≤ 0.06 µg/test recommended.
A test is defined as staining a cell sample (100 µL volume) with empirically determined cell numbers (10⁵–10⁸ cells/test).
Careful titration is recommended for optimal assay performance.

Target Information

CD73 (Ecto-5'-nucleotidase, 5'-ribonucleotide phosphohydrolase) catalyzes the conversion of purine 5'-mononucleotides to nucleosides at neutral pH, with AMP as the preferred substrate.
It forms a dimer of two identical 70 kDa subunits anchored to the plasma membrane via GPI linkage.

CD73 serves as a lymphocyte differentiation marker, with deficiencies linked to various immunodeficiency diseases.
Other forms of 5'-nucleotidase exist in cytoplasm and lysosomes, distinguished by substrate affinity and cofactor requirements.
The CD73 gene is localized to 6q14-q21; defects may lead to joint and arterial calcification or intestinal tuberculosis.
Two transcript variants encoding different isoforms have been identified.
