Target Information

Secreted Frizzled-Related Proteins (sFRPs) are a family of glycosylated Wnt antagonists characterized by a conserved cysteine-rich domain homologous to the extracellular domain of Frizzled proteins.
Unlike Frizzled receptors, sFRPs lack transmembrane and intracellular domains, functioning as soluble modulators of Wnt signaling by directly binding Wnt proteins and inhibiting receptor interaction and signaling.

sFRP-4 is broadly expressed in embryonic and adult tissues and acts as:

  • Circulating antiangiogenic factor
  • Proapoptotic factor
  • Inhibitor of insulin secretion
  • Suppressor of tumor growth and metastasis via Wnt pathway disruption

Downregulation or absence of circulating sFRP-4 correlates with progression of cancers such as ovarian, endometrial, prostate, and lung.
Upregulation is associated with impaired glucose metabolism in type 2 diabetes and suppression of keratinocyte hyperproliferation in psoriasis.
