Target Information
Amphiregulin (AR) binds to EGF-Receptor (EGFR) with lower affinity than EGF. The mature secreted form of AR is an 84-amino acid glycosylated polypeptide growth regulator, generated by proteolytic processing of a 252-amino acid transmembrane precursor.
Seven different polypeptide ligands derived from distinct genes can bind to the extracellular domain of EGFR: EGF, TGFα, AR, HB-EGF, cripto-1, epiregulin, and betacellulin.
All these growth factors contain an EGF-like domain characterized by six evenly spaced cysteine residues forming three disulfide-linked loops.
AR protein is localized in the epithelium of colon, stomach, pancreas, breast, and placenta, and is reportedly overexpressed in human cancers of breast, colon, stomach, and pancreas.
