Target Information

Scn10a (NaV1.8) is a tetrodotoxin-resistant voltage-gated sodium channel alpha subunit. It mediates voltage-dependent sodium ion permeability in excitable membranes and plays a role in neuropathic pain mechanisms. Members of the Deg/ENaC superfamily share structural similarities, including two short intracellular termini, two membrane-spanning segments, and a large extracellular loop with conserved cysteine-rich regions. ENaC proteins (α, β, γ isoforms) regulate sodium and fluid balance in kidney, lung, and colon and are involved in aldosterone-dependent sodium reabsorption and blood pressure control. Mutations in SCN10A are associated with Episodic Pain Syndrome, Familial 2, and Sodium Channelopathy-Related Small Fiber Neuropathy.
