Product Specific Information
Immunohistochemistry (PFA fixed): incubate antibody for 30 minutes at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
BMI1 is a component of the Polycomb group (PcG) multiprotein PRC1 complex, which maintains transcriptional repression of various genes, including Hox genes, throughout development.
The PcG PRC1 complex functions via chromatin remodeling and histone modification, mediating monoubiquitination of histone H2A at Lys-119, resulting in heritable changes in chromatin expressibility.
Within the PRC1 complex, BMI1 is required to stimulate the E3 ubiquitin-protein ligase activity of RNF2/RING2.
