Target Information
RIP1 (Receptor-interacting serine/threonine-protein kinase 1) is a kinase that transduces inflammatory and cell-death signals following activation of death receptors, PRRs, or DNA damage. Upon TNFR1 activation by TNF-alpha cytokines, TRADD and TRAF2 are recruited, leading to phosphorylation of DAB2IP at Ser-728 and activation of the MAP3K5-JNK apoptotic cascade. Ubiquitination via Lys-63-linked chains by TRAF2 enhances signaling through MAPK and NF-κB pathways, promoting inflammatory gene activation. Polyubiquitinated RIP1 binds to IKBKG/NEMO, critical for NF-κB activation. RIPK1-RIPK3 complexes induce programmed necrosis.
