Target Information

Estrogen Receptor beta (ER beta, NR3A2) protein is approximately 55 kDa, though multiple isoforms ranging from 36–59 kDa have been described. Human ER beta was initially cloned and characterized from testis.
Estrogen Receptors (ER) are members of the steroid/thyroid hormone receptor superfamily of nuclear receptors. The estrogen receptor is a ligand-activated transcription factor that, when bound to estrogen, induces a conformational change allowing dimerization and binding to estrogen response elements (ERE) in DNA.
ER can regulate gene transcription positively or negatively through recruitment of coactivator or corepressor proteins.
Two forms of the estrogen receptor exist—alpha (ESR1) and beta (ESR2)—encoded by separate genes. ER beta binds estrogens with an affinity similar to ER alpha and can inhibit ER alpha activity in reproductive tissues.
ER beta is widely expressed in many tissues and may act as a tumor suppressor, playing roles in diseases such as prostate cancer, autism spectrum disorder, medullary thyroid carcinoma, De Quervain disease, and cardiovascular disease risk.
