Target Information
Caspase-activated DNase (CAD) cleaves chromosomal DNA during apoptosis. Apoptosis, a cell death process removing toxic or useless cells during mammalian development, can be induced by various stimuli, including tumor necrosis factor family death factors, UV irradiation, anticancer drugs, and survival factor deprivation.
CAD exists as a complex with its inhibitor, ICAD, which assists proper folding of CAD and inhibits its DNase activity. During apoptosis, caspase activation cleaves ICAD, releasing CAD to enter the nucleus and degrade chromosomal DNA.
Apoptotic signaling is regulated by the balance of pro- and anti-apoptotic proteins (Bcl-2 family), heat shock proteins, and inhibitors of apoptosis proteins (IAPs). Fourteen caspase family members have been identified, seven of which mediate apoptosis.
