Their levels are regulated by synthesis via nucleotide cyclases and degradation by cyclic nucleotide phosphodiesterases (PDEs). PDE6 acts as the effector enzyme in the G protein-mediated visual transduction cascade.
PDE6 consists of multiple subunits:
- Catalytic subunits: alpha′ (Cone), alpha (Rod), and beta (Rod)
- Inhibitory subunit: gamma
- Subunit delta: of unknown function, possibly interacting with other proteins
The catalytic core forms:
- Alpha′/alpha′ homodimers in cones
- Alpha/beta heterodimers in rods
C-terminal modifications such as methylation, myristoylation, and prenylation are essential for proper complex assembly and membrane association.
