Target Information
cAMP is a signaling molecule important for a variety of cellular functions. It activates the cAMP-dependent protein kinase, which transduces signals through phosphorylation of target proteins. The inactive kinase holoenzyme consists of two regulatory and two catalytic subunits. Upon cAMP binding, the holoenzyme dissociates into a regulatory dimer bound to four cAMP and two free catalytic monomers.
In humans, four regulatory and three catalytic subunits have been identified. The PRKAR1A gene encodes one of the regulatory subunits and acts as a tissue-specific extinguisher, down-regulating liver gene expression in hepatoma × fibroblast hybrids. Mutations in PRKAR1A cause Carney complex (CNC). This gene can fuse with the RET proto-oncogene to form the thyroid tumor-specific chimeric oncogene PTC2.
A nonconventional nuclear localization sequence (NLS) suggests a role in DNA replication by serving as a nuclear transport protein for the second subunit of Replication Factor C (RFC40). Three alternatively spliced transcript variants encoding the same protein have been observed.
