Target Information

DNA methylation, the addition of methyl groups to cytosine bases in CpG dinucleotides, is crucial for development and gene regulation. MBD4 is a mammalian demethylase protein containing a methyl-CpG-binding domain (MBD), capable of reverting methylated cytosine bases to cytosine by cleaving the methyl group as methanol. MBD proteins such as MeCP2, MBD1, MBD2, and MBD4 mediate transcriptional repression and DNA methylation signaling. MBD4 also shares homology with bacterial base excision repair DNA N-glycosylases/lyases and is sometimes mutated in microsatellite unstable tumors.
