Target Information
Monomeric G protein Ras acts as a molecular switch linking receptor and nonreceptor tyrosine kinase activation to downstream signaling pathways that regulate cytoplasmic and nuclear events. Mammalian cells contain three Ras proto-oncogenes coding for H-, K-, and N-Ras proteins. Oncogenic mutations in Ras genes occur in approximately 30% of human cancers. Constitutive activation of Ras through mutations or overexpression promotes proliferation and inhibits apoptosis.
- K-Ras mutations: Common in pancreatic, colorectal, and non-small-cell lung carcinomas
- H-Ras mutations: Found in bladder, kidney, and thyroid carcinomas
- N-Ras mutations: Associated with melanoma, hematological malignancies, and hepatocellular carcinoma
