Target Information

ICAD (Inhibitor of Caspase-Activated DNase) is a substrate that regulates nuclear apoptosis by inhibiting caspase-3-activated DNase (CAD).
It exists in two isoforms:

• ICAD-L/DFF45 (45 kDa): functional isoform
• ICAD-S/DFF35 (35 kDa): non-functional isoform

Both isoforms bind CAD, but only ICAD-L is functional. ICAD is cleaved during apoptosis, releasing CAD to induce DNA fragmentation. In non-apoptotic cells, CAD remains complexed with ICAD.
Activation of the CAD/ICAD complex occurs through caspase-3-mediated cleavage at residues 117 and 224, generating three ICAD fragments. Beyond DNase inhibition, ICAD also functions as a CAD-specific folding chaperone.
Recent studies suggest ICAD as a potential therapeutic target for restoring apoptotic signaling in colon and brain cancer cells. Two alternatively spliced transcript variants encode distinct isoforms of ICAD.
