Target Information

ATF6 is a transcription factor that acts during endoplasmic reticulum (ER) stress by activating unfolded protein response (UPR) target genes. It binds DNA on the 5'-CCAC[GA]-3' half of the ER stress response element (ERSE) and ERSE II sequences. Binding to ERSE requires NF-Y interaction. ATF6 forms homodimers and heterodimers with ATF6 beta and interacts with transcription factors such as NF-Y, GTF2I, YY1, and SRF. Under ER stress, the cleaved N-terminal cytoplasmic domain translocates into the nucleus, functioning as a transcription factor. The basic leucine-zipper domain mediates association with NF-Y and DNA binding. During the UPR, a ~50 kDa fragment containing the transcriptional domain is released by proteolysis involving site-1 and site-2 proteases. ATF6 is N-glycosylated and can be phosphorylated by MAPK14/P38MAPK, belonging to the bZIP family.
