• Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
• Formalin-fixed tissues require heating in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at room temperature for 20 minutes.
• This antibody shows broad species reactivity.

Target Information

PGP9.5 (Protein gene product 9.5, UCH-L1, PARK5) is a neuron-specific protein, structurally and immunologically distinct from neuron-specific enolase.
It has a molecular weight of 27 kDa and belongs to the ubiquitin carboxyl-terminal hydrolase family 1 (peptidase family C12).
PGP9.5 exhibits both ubiquitin hydrolase and ligase activities, hydrolyzing small C-terminal adducts of ubiquitin to generate ubiquitin monomers.

PGP9.5 is present in:

• Neurons and nerve fibers throughout the central and peripheral nervous system
• Neuroendocrine cells
• Renal tubules
• Spermatogonia and Leydig cells of the testis
• Ova and corpus luteum cells

Altered expression of PGP9.5 is associated with:

• Overexpression: non-small cell lung cancer
• Decreased expression: Huntington and Alzheimer diseases

It serves as a useful neuronal marker and is valuable for studies of neurodegenerative disorders such as Parkinson disease.
