• IHC (PFA fixed): Incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
• This antibody shows broad species reactivity.

PGP9.5 is a neuron-specific protein distinct from neuron-specific enolase, with a molecular weight of 27 kDa. It belongs to the ubiquitin carboxyl-terminal hydrolase family (peptidase family C12) and exhibits both hydrolase and ligase activities, generating ubiquitin monomers from small C-terminal adducts.

PGP9.5 is expressed in neurons and nerve fibers throughout the central and peripheral nervous systems, as well as in neuroendocrine cells, renal tubules, spermatogonia, Leydig cells, and ovarian tissues.
Overexpression is associated with non-small cell lung cancer, while decreased expression correlates with Huntington’s and Alzheimer’s diseases.
Due to its presence in neuronal inclusions, PGP9.5 serves as a useful neuronal marker and is valuable in neurodegenerative disease studies such as Parkinson’s disease.
