Product Specific Information

This 4NIT4KK monoclonal antibody recognizes human and mouse p38 MAPK when phosphorylated on T180/Y182.
p38 MAPK is a conserved serine/threonine kinase family activated by stress stimuli such as TLR ligands (LPS), osmotic or heat shock, UV irradiation, and inflammatory cytokines.
Members include p38α, β, γ, and δ, activated primarily by MKK3/4 and MKK6.
Downstream targets include MK2/3, p53, ATF-2, and ETS1.
Inhibitor SB203580 can negatively regulate p38 MAPK activity.
Specificity was validated by ELISA, flow cytometry, and western blotting.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated for intracellular staining of human peripheral blood cells at 5 µL (0.06 µg) per test.
A test corresponds to staining a cell sample in a final 100 µL volume (typically 10⁵–10⁸ cells/test).


Staining Protocols

  • Protocol A (Two-step, cytoplasmic): For detection of surface and intracellular (cytoplasmic) proteins.
  • Protocol B (One-step, nuclear): For transcription factors with surface and phosphorylated intracellular proteins.
  • Protocol C (Two-step, fixation/methanol): For optimal phospho-specific signaling discrimination; limited surface staining compatibility.

Target Information

The p38 MAPK cascade regulates cellular responses to stress and inflammation.
Inactive in its nonphosphorylated form, p38 MAPK becomes rapidly activated via dual phosphorylation at Thr180 and Tyr182 by MAPK kinase-3/6.
Activated phospho-p38 MAPK phosphorylates transcription factors such as ATF-2, CHOP-1, and MEF-2, mediating downstream signaling pathways.
