Target Information

DR5 (Apo2, TRAIL-R2, TRICK2, KILLER) is a death domain-containing receptor for TRAIL that mediates TRAIL-induced apoptosis. It belongs to the TNF-receptor superfamily and contains an intracellular death domain. DR5 activation by TNFSF10/TRAIL/APO-2L transduces apoptosis signals. FADD-deficient mouse studies suggest FADD is required for DR5-mediated apoptosis. Two transcript variants encoding different isoforms and one non-coding transcript have been identified. Overexpression of DR5 induces apoptosis and activates NF-κB. DR5 shows high expression in lymphocytes and spleen. Dysfunction of DR5 is associated with squamous cell carcinoma and diffuse infiltrative lymphocytosis syndrome.
