Target Information
Wilms’ tumor (WT) is an embryonal malignancy of the kidney affecting approximately 1 in 10,000 infants, occurring in both sporadic and inherited forms.
The Wilms’ tumor protein (WT1) binds the DNA sequence GCGGGGGCG, a motif common to Egr family Zn²⁺ finger transcriptional activators, and acts as a transcriptional repressor.
WTAP (Wilms tumor 1-associating protein) is a ubiquitously expressed nuclear protein that interacts with WT1 and may regulate mRNA splicing.
WTAP localizes to nuclear speckles and controls the G2/M cell cycle transition by stabilizing cyclin A2 mRNA through binding to its 3′ UTR.
Additionally, WTAP inhibits WT1 target gene expression and impairs WT1 DNA binding ability.
Two isoforms of WTAP exist due to alternative splicing events.
