Product Specific Information

The SK7 monoclonal antibody reacts with human and chimpanzee CD3e, a 20 kDa subunit of the TCR complex. The epsilon chain, along with gamma and delta, is required for proper TCR assembly and surface expression. CD3 is expressed by thymocytes and mature T cells. SK7 and UCHT1 clones cross-block binding, indicating overlapping epitopes.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and validated for flow cytometry of normal human peripheral blood cells. Recommended usage: 5 µL (0.5 µg) per 100 µL test volume.

Super Bright 780 is a tandem dye excited by the violet laser (405 nm) and emits at 780 nm. Recommended filter: 780/60 bandpass. Ensure instrument compatibility.

Compensation values may vary when using UltraComp eBeads (Product #01-2222-42) or AbC Total Antibody Compensation beads (Product #A10497). For best results, set compensation using cells.

When using multiple Super Bright-conjugated antibodies, use Super Bright Complete Staining Buffer (Product #SB-4401) to minimize non-specific polymer interactions.

Light Sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation: Samples can be stored up to 3 days in the dark at 4°C using IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) with minimal brightness loss.

Target Information

The CD3 complex transduces antigen-recognition signals into T cells and regulates TCR expression. It includes CD3 gamma, delta, epsilon, and zeta subunits, encoded on chromosome 11. These subunits have cytoplasmic tails with signal transduction motifs. CD3 is expressed on 68–82% of peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells, but not on B or NK cells. Reduced T cell percentages may occur in autoimmune diseases. Defects in CD3 genes are linked to CD3 immunodeficiency.
