Product Specific Information

The RPA-2.10 monoclonal antibody reacts with human CD2, a 50 kDa cell surface receptor expressed by most thymocytes, all mature T cells, and a subset of NK cells. CD2 interacts with CD58 and is involved in T-cell adhesion and activation. RPA-2.10 blocks mixed lymphocyte reactions and cross-reacts with non-human primates and pigs.

Applications Tested: Pre-diluted and tested on normal human peripheral blood cells at 5 µL (0.06 µg)/test.

Super Bright 780 is a tandem dye excited by the violet laser (405 nm) and emits at 780 nm. Recommended filter: 780/60 bandpass. Ensure instrument compatibility.

Compensation Note: Higher compensation values may occur in the violet 450/50 channel when using UltraComp eBeads microspheres ([Product # 01-2222-42]) compared to single-color stained cells. In such cases, set compensation using cells. Similar effects may occur with AbC Total Antibody Compensation beads ([Product # A10497]).

Staining Panel Consideration: When using multiple Super Bright dyes, use Super Bright Complete Staining Buffer ([Product # SB-4401]) to minimize non-specific interactions.

Light Sensitivity: Protect from light to prevent photo-induced oxidation.

Fixation: Samples may be stored up to 3 days at 4°C in the dark using IC Fixation Buffer ([Product # 00-8222-49]) or 1-step Fix/Lyse Solution ([Product # 00-5333-54]) with minimal brightness loss.

Target Information

CD2 (LFA-2) is a monomeric surface antigen (MW 45–58 kDa) expressed on all peripheral blood T cells and some NK cells, but not on B lymphocytes. It is one of the earliest T-cell markers, present on over 95% of thymocytes. CD2 interacts with CD58 to stabilize T-cell adhesion to antigen-presenting cells. Increased CD2 expression after activation enhances adhesion and immune response. CD2 deficiency can reduce intestinal inflammation and aid infection control. Related diseases include penile squamous cell carcinoma and immune deficiency due to thymus absence.
