Product Specific Information

The SK7 monoclonal antibody reacts with human and chimpanzee CD3e, a 20 kDa subunit of the TCR complex. CD3e, along with gamma and delta subunits, is essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes and all mature T cells.

The SK7 and UCHT1 monoclonal antibodies cross-block binding, suggesting recognition of overlapping epitopes, while OKT3 and SK7 recognize distinct epitopes. SK7 also recognizes chimpanzee CD3.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and validated for flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (0.125 µg) per test (final volume 100 µL, 10⁵–10⁸ cells/test).
Excitation: 633–647 nm; Emission: 660 nm; Laser: Red Laser.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

The CD3 subunit complex is crucial for transducing antigen-recognition signals into T cells and regulating TCR complex expression. CD3 subunits (gamma, delta, epsilon, zeta) are members of the immunoglobulin superfamily encoded on chromosome 11. These subunits associate with signal transduction molecules via tyrosine-based motifs.

CD3 is expressed on 68–82% of normal peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells, but not on B or NK cells. Decreased T lymphocyte percentages may occur in autoimmune diseases. Defects in CD3 genes are associated with CD3 immunodeficiency.
