Product Specific Information

  • Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
  • Staining of formalin-fixed tissues requires heating sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, followed by cooling at room temperature for 20 min.

Target Information

CDC34 accepts ubiquitin from the E1 complex and catalyzes its covalent attachment to other proteins.
It catalyzes Lys-48–linked polyubiquitination and cooperates with E2 UBCH5C and the SCF (FBXW11) E3 ligase complex for polyubiquitination of NFKBIA, leading to its proteasomal degradation.
CDC34 also regulates cell proliferation through ubiquitination and degradation of MYBL2 and KIP1, and targets WEE1 kinase for degradation, influencing the G2/M phase of the cell cycle.
Additionally, it is involved in degradation of β-catenin and interacts dynamically with proteasomes under the influence of human herpes virus 1 protein ICP0.
