Product Specific Information

This 3G8 monoclonal antibody reacts with human and non-human primate CD16, also known as low-affinity Fc gamma RIII.
CD16 exists as two isoforms, FcγRIIIA and FcγRIIIB. FcγRIIIA is expressed on monocytes, macrophages, and NK cells, while FcγRIIIB is found on neutrophils. Expression patterns vary among primate species.

Binding of IgG to CD16 triggers signal transduction pathways leading to ADCC, phagocytosis, cytokine release, and antigen presentation.

Based on cross-blocking studies, 3G8 recognizes the same epitope as CB16 but distinct from B73.1.
Each product contains one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested by flow cytometry of normal human peripheral blood cells.
Use at 5 µL (0.1 µg)/test in 100 µL final volume with 10⁵–10⁸ cells/test.

Important Notes:

  • NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
  • CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
    • Add 5 µL per sample to antibody cocktails.
    • For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton technology with engineered nucleic acid dye structures for optimized fluorescence signatures minimizing spillover and spread.

Target Information

CD16 encodes a receptor that binds the Fc portion of IgG, mediating immune complex clearance and cell-mediated cytotoxicity.
The FCGR3A gene (chromosome 1) encodes the NK cell receptor anchored via transmembrane peptide, while FCGR3B encodes a neutrophil receptor anchored via PI linkage.
Mutations in FCGR3A are associated with Immunodeficiency 20, Herpes Zoster, and susceptibility to lupus and recurrent viral infections.
Alternative splicing produces multiple isoforms.
