Product Specific Information

The 2D1 monoclonal antibody reacts with all isoforms of human CD45 (Leukocyte Common Antigen, LCA).
CD45 is expressed by all hematopoietic cells excluding circulating erythrocytes and platelets.
Its cytoplasmic region exhibits tyrosine phosphatase activity, crucial for lymphocyte activation.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells at 5 µL (0.8 µg) per test.
A test is defined as staining a cell sample of 100 µL volume (10⁵–10⁸ cells/test).

Compatibility Notes:
NovaFluor dyes are incompatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI.
Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.

CellBlox Plus Blocking Buffer:
Updated conjugates ship with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01).
Add 5 µL per sample for optimal staining.
For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD45 (LCA) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells (~10% of lymphocyte surface proteins).
Absent in non-hematopoietic tissues, CD45 regulates Src-family kinases, T/B-cell receptor signaling, and cytokine receptor pathways.
Multiple isoforms arise from alternative splicing, differing in extracellular domains.
CD45 is essential for lymphocyte activation, survival, and immune regulation.
