This 23dcdp monoclonal antibody reacts with the p19 subunit of human IL-23, a heterodimeric cytokine composed of p40 and p19 subunits. IL-23 shares the p40 subunit with IL-12 and signals through a heterodimeric receptor (IL-12Rβ1 and IL-23R). IL-23R signaling activates the Jak/STAT pathway and induces RORγt expression, promoting Th17 cell maintenance and proliferation.

Dendritic cells and macrophages produce p40 and IL-23 in response to TLR2, TLR4, TLR8 ligands, and Dectin-1 agonists. Expression levels vary depending on donor, kinetics, and stimuli type. The p19 subunit may also exist independently of p40 association.

• Pre-titrated and validated for intracellular staining and flow cytometry of cultured human dendritic cells.

Target Information

IL-23 is a heterodimeric cytokine composed of the p40 subunit (shared with IL-12) and p19. While p19 alone is inactive, IL-23 interacts with IL-12Rβ1 and IL-23R to signal through STAT4. Secreted by activated dendritic cells, IL-23 promotes proliferation of memory T cells and induces IL-17 production, distinguishing its function from IL-12, which primarily induces IFN-γ.

Human IL-23 similarly promotes memory T cell proliferation and moderate IFN-γ production compared to IL-12.
