Target Information
Mutations in the ABCA1 gene (ATP-binding cassette transporter 1) are associated with Tangier disease (TD), an autosomal recessive disorder characterized by an absence of plasma HDL, accumulation of cholesterol esters in the reticuloendothelial system, and altered cellular lipid trafficking.
ABCA1 mediates the apoA-I–dependent export of cholesterol and phospholipids from cells. It is expressed on the plasma membrane and Golgi complex and is regulated by cholesterol flux. The balance of cholesterol flux between HDL and macrophages is competitively regulated by ABCA1 and SR-BI.
