Target Information

EIF2AK2 (PKR) is one of four kinases that specifically phosphorylate Ser51 of translation initiation factor eIF2-alpha in response to various environmental stresses. PKR plays a key role in antiviral defense through activation by double-stranded RNA produced during viral infection. It is involved in IFN-induced antiviral response, virus-induced apoptosis, and regulation of cell growth and differentiation.

PKR induces apoptosis by up-regulating Fas expression and mediates the FADD/Caspase-8 death signaling pathway. Upon viral infection, PKR functions as a dual protein, activating both cell survival and death pathways through kinase-dependent and independent mechanisms. It regulates multiple pathways including NF-kappaB activation, p53, p38, and PDGF signaling. PKR has been implicated in tumor suppression and malignancy, as well as in neurodegenerative diseases such as Alzheimer’s, Huntington’s, and amyotrophic lateral sclerosis. Viruses have evolved mechanisms to evade PKR, such as inhibition by influenza virus NS1 protein.
