Target Information
Thioredoxins (Trx) are small, multifunctional proteins with oxidoreductase activity and are ubiquitous in living cells. They contain a conserved Cys-Gly-Pro-Cys active site forming redox-active disulfide/dithiol groups. The active site disulfide can be reduced by thioredoxin reductase using NADPH as an electron donor. Reduced Trx serves as a hydrogen donor for ribonucleotide reductase, essential for DNA synthesis, and functions as a general protein disulfide reductase involved in growth and redox regulation. Trx reduces targets such as protein disulfide-isomerase (PDI) and transcription factors including p53, NF-κB, and AP-1. It also contributes to H₂O₂ removal when coupled with methionine sulfoxide reductase or peroxiredoxin isoforms.
