Product Specific Information

The 11B11 antibody reacts with mouse interleukin-4 (IL-4), a 14 kD cytokine, and functions as a neutralizing antibody. Mouse IL-4 (also known as BCDF, BCGF-1, BSF-1) is secreted primarily by activated Th2 and NKT cells, and to a lesser extent by Th1 and mast cells. IL-4 promotes B cell proliferation, differentiation, and Ig isotype switching, and upregulates surface MHC class II and CD23.

Applications Reported:
Suitable for intracellular staining followed by flow cytometric analysis.

Applications Tested:
Tested by intracellular staining and flow cytometry of Th2-polarized, stimulated mouse splenocytes. Recommended use ≤ 0.125 µg/test (defined as staining a 100 µL cell sample). Cell number should be empirically determined (10⁵–10⁸ cells/test). Titration is recommended for optimal performance.

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm and is excited with the blue laser (488 nm). Recommended filter: 710/50 bandpass (695/40 acceptable). Ensure instrument compatibility.

Fixation:
Samples can be stored in:

• 1-step Fix/Lyse Solution (Product #00-5333-54)

Storage up to 3 days at 4°C in the dark with minimal signal loss. Clone-specific fixation performance should be empirically verified.

Target Information

Interleukin-4 (IL-4) is a pleiotropic cytokine produced by activated T cells and acts as a ligand for the IL-4 receptor. It shares receptor components with IL-13, leading to overlapping biological functions. STAT6 is a key mediator of IL-4 signaling. The IL-4 gene is part of a cytokine cluster on chromosome 5q, closely located to IL-13 and IL-5, regulated by long-range genomic elements. IL-4 delta 2 is a known splice variant.
