Product Specific Information

This fc23cpg monoclonal antibody reacts with the p19 subunit of mouse IL-23, a heterodimeric cytokine composed of p40 and p19 subunits. IL-23 shares the p40 subunit with IL-12 but signals through a distinct receptor complex (IL-23R/IL-12Rβ1), activating the Jak/STAT pathway.
Unlike IL-12, IL-23 supports Th17 differentiation via RORγt expression. Dendritic cells and macrophages produce IL-23 in response to TLR2, TLR4, TLR8 ligands, and Dectin-1 agonists.

• Applications Reported: Intracellular staining followed by flow cytometric analysis.
• Applications Tested: Tested on stimulated mouse bone marrow-derived dendritic cells using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00).
  Use ≤0.125 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Optimize antibody concentration empirically.

Fluorochrome Information:
PerCP-eFluor® 710 emits at 710 nm (excited at 488 nm). Use a 710/50 or 695/40 bandpass filter. Protect from light due to photo-induced oxidation sensitivity.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222) or 1-step Fix/Lyse Solution (Product #00-5333) for up to 3 days in the dark at 4°C with minimal signal loss. Filtration: 0.2 µm post-manufacturing filtered.

Target Information

IL-23 is a heterodimeric cytokine composed of the IL-12 p40 subunit and a unique p19 subunit. The p19 subunit alone is inactive. IL-23 binds IL-12Rβ1 and IL-23R, activating STAT4-mediated signaling.
It is secreted by activated mouse and human dendritic cells.
Mouse IL-23 induces proliferation of memory T cells and IL-17 production, but not IFN-γ.
Human IL-23 similarly promotes memory T cell proliferation and moderate IFN-γ induction.
