Target Information
Receptor-tyrosine-kinase-like orphan receptor 1 (ROR1) is a tumor-associated surface protein predominantly expressed during embryogenesis, playing roles in organ morphogenesis, nervous system development, and neural progenitor maintenance. It is virtually absent from normal adult tissues except for low-level expression in immature B cell precursors and adipocytes. ROR1 is overexpressed in several malignancies, including chronic lymphocytic leukemia (CLL), mantle cell lymphoma (MCL), acute lymphoblastic leukemia (ALL), marginal zone lymphoma, and lung adenocarcinoma.
Initially identified in neuroblastoma cell lines, ROR1 was classified as an orphan receptor until Wnt-5a was suggested as a potential ligand. ROR1 functions as a pseudokinase, promoting proliferation and resistance to apoptosis through interactions with Wnt-5a and TCL1-mediated AKT activation. Structurally, ROR1 is a glycoprotein with extracellular Ig-like, Frizzled, and Kringle domains, and an intracellular tyrosine kinase domain.
