Product Specific Information

Description:
The MEM-15 monoclonal antibody reacts with human CD14, a 53–55 kDa GPI-linked glycoprotein expressed on monocytes, interfollicular macrophages, and some dendritic cells. Complexes of LPS and LBP (LPS-Binding Protein) bind with high affinity to monocytes via surface CD14.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:

• Flow cytometric analysis

Applications Tested:

• Pre-diluted and validated for flow cytometric analysis of normal human peripheral blood cells.
• Recommended use: 5 µL (0.05 µg)/test in a final volume of 100 µL.
• Cell number: 10⁵–10⁸ cells/test (empirically determined).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL per sample to antibody cocktails before mixing with cells.
• For single-color controls, use 5 µL of CellBlox per 100 µL of cell sample (10³–10⁸ cells).

Technology:
NovaFluor conjugates utilize Phiton technology with engineered nucleic acid dye structures, minimizing spillover and spread.

Target Information

CD14 is a 55 kDa GPI-anchored glycoprotein expressed on monocytes, macrophages, and neutrophils. It functions as a multifunctional lipopolysaccharide receptor and exists in both membrane-bound and soluble forms. CD14 binds LPS in conjunction with LBP, playing a role in immune recognition and inflammatory processes. Soluble CD14 (sCD14) can distinguish structural variations in LPS and contributes to neutralization of serum endotoxins. CD14 also binds apoptotic cells and is implicated in allergic, inflammatory, and infectious responses.
Associated diseases include Mycobacterium chelonae infection and croup.
