Product Specific Information
The MEM-15 monoclonal antibody reacts with human CD14, a 53–55 kDa GPI-linked glycoprotein expressed on monocytes, interfollicular macrophages, and some dendritic cells. Complexes of LPS and LBP bind with high affinity to monocytes through surface CD14.
Each product contains:
- 1 vial of NovaFluor conjugate
- 1 vial of CellBlox Plus Blocking Buffer
Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells. Recommended use at 5 µL (0.8 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
- CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
- Add 5 µL per sample prior to combining antibody cocktails with cells.
- For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).
NovaFluor conjugates utilize Phiton™ technology, based on novel nucleic acid dye structures engineered for optimized fluorescent signatures, minimizing spillover and spread effects.

Target Information
CD14 is a 55 kDa GPI-anchored glycoprotein expressed on mature monocytes, macrophages, and neutrophils. It functions as a multifunctional lipopolysaccharide receptor and exists both as a membrane-bound and soluble form. CD14 binds LPS via LBP, and soluble sCD14 can discriminate structural variations between lipopolysaccharides, aiding in neutralization processes. It also binds apoptotic cells and influences allergic, inflammatory, and infectious responses.
Diseases associated with CD14 dysfunction include Mycobacterium chelonae infection and croup.
Alternative splicing results in multiple transcript variants encoding the same CD14 isoform.
