Product Specific Information

The MEM-15 monoclonal antibody reacts with human CD14, a 53–55 kDa GPI-linked glycoprotein expressed on monocytes, interfollicular macrophages, and some dendritic cells. Complexes of LPS and LBP bind with high affinity to monocytes via CD14.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells at 5 µL (0.6 µg) per test (100 µL final volume). Suitable for 10⁵–10⁸ cells/test.

Compatibility Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

CellBlox Plus Blocking Buffer:
This conjugate ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which provides improved formulation over the original CellBlox. Use 5 µL per sample for optimal staining. For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD14 is a 55 kDa GPI-anchored glycoprotein expressed on mature monocytes, macrophages, and neutrophils. It functions as a multifunctional lipopolysaccharide receptor and exists both as a membrane-bound and soluble form. CD14 binds LPS via LBP, distinguishing structural variations among lipopolysaccharides and aiding in neutralization processes. It also binds apoptotic cells and influences allergic, inflammatory, and infectious responses.
Associated diseases include Mycobacterium chelonae infection and Croup.
