Product Specific Information
The eBioH4A3 monoclonal antibody reacts with human CD107a, also known as lysosomal-associated membrane protein-1 (LAMP-1). CD107a is a highly glycosylated ~110 kDa protein expressed mainly in lysosomal/endosomal membranes across most cell types. It is transiently expressed on the surface of degranulating cytolytic T cells and is upregulated on activated platelets and certain cancer cells.

- Surface staining in flow cytometric-based degranulation assays

- Pre-titrated and tested by intracellular staining and flow cytometric analysis of normal human peripheral blood cells

Target Information
LAMP1 (CD107a, lysosome-associated membrane protein-1) is a major lysosomal membrane component. Approximately 1–2% of total CD107a is also found on the plasma membrane. It contains a signal peptide, 18 N-linked glycosylation sites, a single membrane-spanning region, and a short cytosolic tail.
LAMP proteins are essential for lysosome biogenesis and lysosome-phagosome fusion. Upon activation, LAMP1 translocates to the plasma membrane via a tyrosine-based motif (YXXI). Disruption of this motif prevents lysosomal localization.
Surface LAMP1 promotes adhesion of PBMCs to vascular endothelium, suggesting a role in inflammation. Increased LAMP1 expression is observed in neurons and glial cells near senile plaques in Alzheimer’s disease and in macrophages and lymphocytes during thymic involution. LAMP1 serves as a marker for mast cell activation.
