Target Information
TID1은 Drosophila tumor suppressor lethal (2) tumorous imaginal discs, l(2)tid의 인간 상동체로, 두 가지 미토콘드리아 매트릭스 국소화 스플라이스 변형체(hTid-1S 및 hTid-1L)를 암호화합니다.
이 단백질들은 DnaJ 단백질 계열의 보존된 구성원으로, 미토콘드리아 Hsp70의 코샤페론으로 작용합니다.
J 도메인은 Hsp70 샤페론에 결합하여 ATPase 활성을 활성화합니다.
hTid-1L의 발현은 DNA 손상 유도제(예: mitomycin-C, TNF-α)에 의해 유도된 세포사멸을 증가시키며, J-도메인 변이체는 세포사멸을 억제할 수 있습니다.
반면 hTid-1S의 J-도메인 변이체는 세포사멸을 증가시킵니다.
이 두 스플라이스 변형체는 미토콘드리아에서 cytochrome c 방출과 caspase 3 활성화를 조절하며, caspase 8 활성화에는 영향을 주지 않습니다.
이들의 상대적 발현량에 따라 미토콘드리아가 세포사멸 신호를 조절할 수 있는 것으로 제안됩니다.
