Target Information
MMP(matrix metalloproteinase)는 결합 조직 성분을 분해할 수 있는 단백질분해 효소로, 금속 결합 활성 부위의 보존된 아미노산 서열을 가지며, TIMP(tissue inhibitors of metalloproteinases)에 의해 억제됩니다.
MMP와 TIMP는 혈관신생 조절에 중요한 역할을 합니다.
MMP2는 631개의 아미노산으로 구성된 전구 효소 형태로 합성되며, 처음 80개의 아미노산 절단을 통해 활성화됩니다. 구조적으로는 프로펩타이드, 촉매, 헴오펙신 도메인을 포함합니다.
MMPs는 최소 18종 이상의 분비형 및 막결합형 아연 엔도펩티다아제 패밀리로, 세포외기질의 모든 구성 요소(콜라겐, 피브로넥틴, 라미닌 등)를 분해할 수 있습니다. 일반적으로 신호 펩타이드, 프로펩타이드, 그리고 보존된 아연 결합 부위를 포함하는 촉매 도메인을 가지고 있습니다.
기능적으로 MMP2는 조직 재형성에 관여하며, MMP2 유전자 변이는 Winchester 증후군 및 Nodulosis-Arthropathy-Osteolysis(NAO) 증후군과 관련이 있습니다. 두 가지 전사체 변형으로 서로 다른 MMP2 이소형이 존재합니다.
