Target Information
Matrix metalloproteinases (MMPs) are proteolytic enzymes capable of degrading connective tissue components. They share a common activation mechanism, a conserved amino acid sequence in the metal-binding active site region, and are inhibited by tissue inhibitors of metalloproteinases (TIMPs).
MMPs and TIMPs regulate angiogenesis and tissue remodeling. MMP2 is synthesized as a 631 amino acid proenzyme that becomes active upon cleavage of the first 80 amino acids. It contains propeptide, catalytic, and hemopexin domains.
MMP2 mutations have been associated with Winchester syndrome and Nodulosis-Arthropathy-Osteolysis (NAO) syndrome. Two transcript variants encoding different isoforms of MMP2 have been identified.
