Product Specific Information
- Heat-mediated antigen retrieval using Tris/EDTA (pH 8) is recommended for paraffin sections.
- The epitope recognized by clone 514H12 is sensitive to peroxide; perform peroxidase-blocking steps after antibody incubation.
- Suggested positive control: human tonsil tissue.
- Mouse anti-human CD68 antibody (clone 514H12) recognizes a ~110 kDa glycoprotein primarily expressed by macrophages and monocytes.
Target Information
CD68 (Macrosialin) is a 110 kDa integral membrane glycoprotein predominantly expressed on intracellular lysosomes of monocytes and macrophages, and to a lesser extent by dendritic cells and granulocytes.
It plays roles in phagocytic activity, lysosomal metabolism, and cell-cell or cell-pathogen interactions.
CD68 is expressed by interdigitating reticulum cells in tonsil, histiocytic lymphoma, histiocytosis, acute myeloid leukemia (AML), and granulocytic sarcoma.
Elevated CD68 expression has been observed in CD34+ cells in various human malignancies, including AML studies.
