Description:
The HIT3a monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. Together with CD3γ and CD3δ, the ε chain is essential for proper assembly, trafficking, and surface expression of the TCR complex. CD3 is expressed by thymocytes in a developmentally regulated manner and by all mature T cells. Crosslinking of TCR with HIT3a initiates intracellular signaling leading to cellular activation and proliferation.

Applications Reported:
The HIT3a antibody has been reported for use in flow cytometric analysis.

Applications Tested:
The antibody has been tested by flow cytometric analysis of normal human peripheral blood cells. Recommended use is ≤ 0.25 µg per test (100 µL final volume). Cell number may range from 10⁵ to 10⁸ cells/test. Antibody should be titrated for optimal performance.

Purity: Greater than 90%, as determined by SDS-PAGE.
Aggregation: Less than 10%, as determined by HPLC.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

The CD3 subunit complex is crucial in transducing antigen-recognition signals into the cytoplasm of T cells and in regulating TCR complex surface expression. CD3 subunits (γ, δ, ε, ζ) are structurally related members of the immunoglobulin superfamily encoded on human chromosome 11. These subunits contain cytoplasmic tails with double tyrosine-based motifs that mediate signal transduction. CD3 is expressed on 68–82% of peripheral blood lymphocytes, 65–85% of thymocytes, and Purkinje cells in the cerebellum, but not on B or NK cells. Decreased T lymphocyte percentages may occur in autoimmune diseases. Mutations in CD3 genes are associated with CD3 immunodeficiency.
